How does the MTT assay actually work?

MTT is a yellow tetrazolium salt that metabolically active cells reduce, via mitochondrial dehydrogenase enzymes, into purple insoluble formazan crystals. After solubilizing the crystals, the intensity of the resulting color is measured spectrophotometrically and is proportional to the number of viable, metabolically active cells.

What can interfere with MTT assay results?

Compounds that are themselves colored, that directly reduce the tetrazolium salt independent of cellular metabolism, or that interfere with mitochondrial enzyme activity without actually killing the cell can all produce a misleading signal — a known limitation that is accounted for with appropriate controls when testing colored or redox-active test compounds.

Is MTT a measure of cell number, cell viability, or cell proliferation?

MTT signal correlates with the number of metabolically active cells, so it is commonly used as a proxy for all three, but strictly speaking it measures mitochondrial dehydrogenase activity — a change in per-cell metabolic activity (without a change in cell number) can shift the MTT signal independent of actual proliferation or death.

How long does an MTT assay take?

A typical MTT assay involves incubating cells with the test compound, then adding the MTT reagent for a further incubation period (commonly 1-4 hours) to allow formazan formation, followed by solubilization and spectrophotometric reading — the full protocol is usually completed within the same day after the initial compound exposure period.

Why is MTT still widely used despite having several newer alternatives?

MTT remains popular because it is inexpensive, technically simple, requires only a standard spectrophotometer (no specialized luminometer or flow cytometer), and has decades of published reference data across countless cell types and compounds, making results easy to benchmark against the existing literature.
